MES Hydrate (CAS 1266615-59-1) is a zwitterionic biological buffer belonging to the Good's buffer family. It is widely used in biological, biochemical, and environmental research due to its excellent buffering capacity, high water solubility, and minimal interaction with metal ions.

 

With an effective buffering range of pH 5.5–6.7 and a pKa value of 6.1 at 25°C, MES Hydrate provides stable pH control under physiological conditions. Its low UV absorbance, minimal chemical reactivity, and low cytotoxicity make it particularly suitable for sensitive biological applications.

 

Compared with commonly used buffers such as Tris and phosphate, MES Hydrate demonstrates lower toxicity toward cells and reduced metal ion chelation, ensuring greater compatibility with biological systems and metal-sensitive assays.

Cell Culture: Used as a buffering agent in cell culture media to maintain stable pH conditions while minimizing cellular stress.

Protein Purification and Extraction: Incorporated into extraction and purification buffers due to its buffering stability and low metal ion binding.

Enzyme Studies: Applied as a component of extraction buffers for BACE1 and reaction mixtures for measuring 5,10-methenyltetrahydrofolate synthetase (MTHFS) activity in fibroblasts.

Bioconjugation: Employed in MES coupling buffers during the immobilization of monoclonal antibodies (mAbs) and GST-fusion proteins onto carboxylate-modified polystyrene latex (CML) beads.

Fluorescent Labeling: Used to adjust and maintain optimal pH conditions during fluorescence labeling procedures.

Gel Electrophoresis: Commonly used as a running buffer component in denaturing electrophoresis for reliable protein separation and analysis.
